Detection of beverage spoiler using real-time PCR

 

Application:

The detection and identification of beverage spoilage microorganisms has so far been carried out using time-consuming cultural selective enrichment. Only after at least five days does a turbidity of the cultured sample indicate an infestation with beverage spoiler, often too late to be able to take countermeasures in time. The PCR method now offers beverage producers the possibility of reliably detecting contamination with microorganisms at any point in the production process within just a few hours. To prevent the transfer of harmful microorganisms into the product, both the raw materials used and the equipment and pipe systems used should be checked for microbial infestation at regular intervals.

Detection is performed by fluorescence measurement using the hydrolysis probe format (TaqMan®). Using hot-start PCR plus double-labelled sequence-specific probes (FAM / HEX / ROX / CY5 or ATTO 490LS), a measurable fluorescence signal of a defined wavelength is emitted during the extension phase if hybridization to the target sequence is correct. An inhibition control is amplified together with the specific sequence in a reaction tube to exclude false negative results due to inhibition.
