Application:
Manual DNA extraction from drinking water, cooling water, waste water and sewage sludge using silica membrane technology
The background to this is the new Directive (EU) 2020/21842 of the European Parliament and of the Council of 16 December 2020 on the quality of water intended for human consumption with the first-time specification of a parameter value for the Legionella parameter of < 1,000 CFU/L for the newly introduced risk assessment of drinking water installations.
The test applies to hot water systems with a storage tank volume of 400 liters or more or with a pipe volume of more than 3 liters between the water heater and the distribution point.
The test frequency for systemic testing for legionella in a building water supply system is initially once a year when drinking water is supplied to the public. Inspection intervals of up to three years by the public health department are possible.
Principle:
The method is based on spin column technology. After chemical lysis of the cells, the DNA binds to a silica membrane. Cell residues, salts and inhibitors are removed by washing steps. The DNA is released from the silica membrane by heating in TE or H2O. The kit is robust against strongly acidic or alkaline, contaminant-rich and difficult to isolate samples. The DNA is of high purity and is available for further molecular biological analyses (e.g. real-time PCR, sequencing….).
